Rapid PCR, RT-PCR, and Antigen Testing — What’s the Difference?
As COVID-19 testing became part of everyday life, three types of tests dominated the conversation: RT-PCR, Rapid PCR, and Antigen tests. Each has different strengths, limitations, and use cases. Here is a clear guide to understanding the differences.
RT-PCR (Reverse Transcription Polymerase Chain Reaction)
RT-PCR is the gold standard for COVID-19 diagnosis. It detects the genetic material (RNA) of the SARS-CoV-2 virus in a nasal or throat swab sample. The process involves:
Converting viral RNA to DNA (reverse transcription)
Amplifying the DNA millions of times (PCR) to make it detectable
Detecting the amplified DNA using fluorescent probes

Rapid PCR
Rapid PCR uses the same RT-PCR technology but in a miniaturised, automated format that delivers results in 45–90 minutes rather than hours. It is used in point-of-care settings — airports, hospitals, clinics — where fast results are needed. Sensitivity and specificity are comparable to standard RT-PCR.

Rapid Antigen Test (RAT)
Antigen tests detect viral proteins (antigens) rather than genetic material. They are faster and cheaper than PCR tests but less sensitive — meaning they are more likely to miss infections, especially in people with low viral loads (early or late infection, or asymptomatic cases).
